Application:
Biochemical reagents, preparation of biological media, such as the preparation of HAT selective media.
Preparation of HAT medium (for reference only) :
100×A (aminopterin) storage solution: weigh 1.76mg aminopterin dissolved in 90ml ultrapure water, add 0.5ml of 1mol/L NaOH drop to help dissolve, after completely dissolved, add 0.5ml of 1mol/L HCL to neutralize, and then add ultrapure water to 100ml. The bacteria were removed by 0.22um membrane filtration, and small amounts were divided into aliquots and stored at -20 ℃.
100×HT storage solution: Weigh 136.1mg hypoxanthine and 38.8mg thymidine, add ultrapure water to 100ml, place in 45-50℃ water bath to dissolve completely, filter to remove bacteria through 0.22μm membrane, aliquots in small amount, store at -20℃, dissolve in 37℃ water bath before clinical use.
HT medium: 82ml basal medium was supplemented with 1ml HT (100×), 1ml SP (100×), and 15ml inactivated calf serum.
HAT medium: 99ml HT medium was added to 1ml A (100×).
