 /></p><p> Product Introduction: </p><p>MTT can be reduced by some dehydrogenases in the mitochondria to form the crystalline dark purple product formazan, which can be completely dissolved in the presence of a specific solvent, and then the absorbance near the wavelength of 490nm can be measured by an enzymoscope. The more cells proliferate and the faster, the higher the absorbance; The greater the cytotoxicity, the lower the absorbance. </p><p> Instructions for use: </p><p>1. Cells of logarithmic phase were collected and the concentration of cell suspension was adjusted and divided into 96-well plates with 180μL per well and 3000-10000 cells/well. </p><p>2. The cells were cultured at 37℃ and 5%CO2 for 6-24 hours. </p><p>3. Add the appropriate concentration of the tested compound and continue to culture for an appropriate time. </p><p>4. Carefully absorb the supernatant, add 90μL fresh culture solution, then add 10μL MTT solution, and continue to culture for 4 h. </p>5. Then absorb the supernatant, add 110μL Formazan solution to each well, and oscillate at a low speed for 10 min on a shaking table to fully dissolve the crystal. The light absorption values of each hole were measured at 490 nm by enzyme-linked immunoassay. </p><p>6. Zero holes (medium, MTT, Formazan solution) and control holes (cells, drug dissolving medium of the same concentration, culture medium, MTT, Formazan solution) were set up at the same time, and 3 multiple holes were set up in each group. </p><p> Note: </p><p>1. Due to the use of 96-well plates for detection, if the cell culture time is long, we must pay attention to the problem of evaporation. On the one hand, since a circle around the 96-well plate is the easiest to evaporate, the method of abandoning the circle around the plate can be adopted to add PBS, water or culture solution; On the other hand, the 96-well plate can be placed close to the water source in the incubator to ease evaporation. </p><p>2. The MTT solution is yellow and should be kept away from light. Long-term light will cause failure. Do not use when the color turns grayish green. Formazan solution can be incubated in a water bath at 37 ° C to promote dissolution when frozen or precipitated, and must be used after all dissolving and mixing. </p><p>3.
