Polylysine can be used to promote the adherent growth of cells.
Principle:
Polylysine solution is widely used as an adhesive for tissue sections and slides. The interaction between the polycation molecule and the anion on the tissue section will produce a strong adhesion force. The properties of the flask surface are very important for cell culture. The glycoprotein (negative) on the cell surface is easy to adsorb on the hydrophilic surface, so the cell culture surface with a considerable amount of positive charge can promote cell adsorption, which is the importance of optimizing the cell culture surface with poly-L-lysine.
Application:
It is suitable for anti-slide processing of slides used in histology, immunohistochemistry, frozen section, cell smear, in situ hybridization, etc., to prevent tissue slides from falling off during experimental procedures. It can also be used in cell culture to increase cell adhesion ability.
Solution preparation:
It was dissolved in deionized water and prepared into mother liquor for use. It was stored at -20 ℃.
Slide processing:
1. Sterilized water diluted polylysine solution, the general use concentration is 0.01%.
2 Dip the slide in a diluted polylysine solution for 5 minutes. Note that adding time does not improve the effect of the coating.
3. Dry in the oven at 60℃ for 1 hour, or dry at room temperature 18-26℃ overnight for use.
Note:
1. Every 100mL diluted poly-lysine solution should be coated with 40-90 slides, more than 90 slides will affect its adhesion.
2. The glass slides must be kept clean before use. When necessary, a 70% ethanol solution containing 1% HCl was used for cleaning.
3. The released polylysine solution should be placed at 2-8 ℃ and is stable for at least 3 months.
4. The used diluent should be filtered and discarded if it becomes cloudy or grows bacteria
5. The concentration of polylysine can be higher, and the aspirated can be reused for at least 20 times.
Culture bottle treatment:
1. Coated culture plate or culture bottle, generally polylysine concentration is 0.01%,
2. Prepare 100ml of triple distilled water and autoclaving.
3. 0.01% polylysine was uniformly applied to the surface of the culture substrate at an amount of 50 microliters per square centimeter and allowed to stand for 5min at room temperature until excess liquid was removed.
4. Add sterilized water, wash repeatedly three times, aseptic operation.
5. Dry aseptically after treatment for use.
6. The sterile polylysine can be reused after recycling.
