Product Description:
In vitro culture of histiocytic cells, in primary cell culture of histiocytic dispersion (preparation of tissue blocks into a single cell suspension), and in pass-through cell culture, digestion and dispersion of adherent growth cells are performed using histiocytic digestion fluid. Commonly used digestive fluids are trypsin-EDTA, etc., whose function is mainly to hydrolyze intercellular proteins (such as extracellular matrix), disperse tissues or adherent cells into single cells, and make cell suspension for further experiments.
T1350 Trypsin Solution produced by Solarbio contains 0.25% trypsin, does not contain EDTA and phenol red, dissolves in calcium and magnesium free balanced salt solution, and can be directly used for digestion of cultured cells and tissues after filtration. This product has the characteristics of convenience, fast, stable and safe, good cell state, etc. Most adherent cells can be digested in about 2 minutes at room temperature.
Storage: -20℃ storage, valid for 12 months.
Usage:
1. Digestion of adherent cells:
a) Remove the culture solution and wash the cells once with sterile PBS, Hanks solution, or serum-free culture solution to remove the residual serum.
b) Add a small amount of trypsin digestion solution, cover the cells, and leave for 1-2 minutes at room temperature. The digestion time of different cells is different, and the digestion time can be extended appropriately for cells with solid adhesion.
c) the cells contracted significantly under the microscope, and the morphological changes of the cells were found at the bottom of the culture vessel by naked eye; Or you can blow a cell with a gun and find that the cell just blows off. Blot out the digestive juices at this point. Add the cell culture medium containing serum and blow down the cells, which can be directly used in subsequent experiments.
d) If insufficient digestion is found, trypsin digestion solution can be added for re-digestion.
e) If it is found that the digestion time of the cells is too long and the cells are not blown, some of the cells have fallen off directly from the bottom of the culture vessel, and the cells are blown down directly with the pancreatic enzyme cell culture solution. Centrifuge 1000-2000g for 1 minute, precipitate the cells, remove the digestive fluid of pancreatic enzyme cells as much as possible, and then add the complete culture solution containing serum to re-suspend the cells, which can be used for follow-up experiments.
2. Tissue digestion:
The time required for digestion of different tissues varies greatly, and it is usually appropriate to fully break up the tissues after digestion.
Note:
1. Due to the different nature of tissues or cells, the experimental personnel should determine the best digestion time according to the specific situation; The time of digesting cells should not be too long, otherwise it will affect the cell adhesion and growth;
2. This product does not contain bacteriostatic agents, and special attention should be paid to aseptic operation during use to avoid microbial contamination of digestive fluid;
3. It should not be stored at 4℃ for a long time, do not freeze and thaw repeatedly, and it is recommended to pack and freeze when using a small amount.
