It is a substrate of horseradish peroxidase (HRP). ABTS is oxidized to form a stable blue-green cationic ABTS radical with maximum absorption peaks at 405 nm or 734 nm. After the tested substance is added to ABTS free radical solution, the antioxidant components can react with ABTS free radicals and make the reaction system fade, and the absorbance of 405 nm decreases, and the change of absorbance is proportional to the degree of free radical removal within a certain range. The absorbance drop was measured to reflect the ability of the sample to remove ABTS free radicals. ABTS method can be used to determine the antioxidant capacity of hydrophilic and lipophilic substances.


Examples of using this product(for reference only)

ABTS Free Radical Scavenging Assay.

1To be specific, ABTS (25 mg) was added to the potassium persulfate (6.5 mL, 2.45 mmol/L) solution. Subsequently, the prepared ABTS solution was stored (14 h) at a constant temperature (25 ℃).The maintenance of the absorbance (734 nm, 0.70 ± 0.10) was confirmed before the experiments. Then, fresh MPN-GAs (1.0 mL, 1 mg/mL) and the prepared ABTS (3 mL) solution were subjected to an incubation process (60 min). Final ABTS scavenging activities were evaluated through the transformation of measured absorbance (734 nm).

2ABTS was dissolved in 25 mM Tris, pH 7.0, to a 7 mM concentration.

The ABTS radical cation (ABTS+·) was produced by mixing the ABTS stock solution with 2.45 mM potassium persulfate at a ratio of 1:1 (v:v) following 12?16 h of incubation in the dark (4 ℃). The solution was then diluted with Tris buffer to an absorbance of 0.70 (±0.02) at 734 nm to form the test reagent. The test sample (3 μL) was then mixed with 237 μL of the diluted ABTS solution. The mixture reacted against light at room temperature for 30 min, and the absorbance was recorded at 734 nm on a ultraviolet spectrometer (AE). The blank was made from 3 μL of Tris buffer and 237 μL of diluted ABTS solution (AB). The percentage of inhibition of ABTS+· was calculated using the formula:rate of inhibition=[ (AB-AE)/AB ] X 100where AB is the absorbance of the blank sample and AE is the absorbance of the mixtures.
