An important nutrient and oxidizing reductant that plays an important role in bone salt metabolism and bone formation. In Vitro, it can increase calcium salt deposition, promote the formation of mineralized nodules, and indirectly regulate osteoclast differentiation through mesenchymal cells. Vitamin C can directly inhibit osteoclast formation induced by nuclear factor κB receptor activator ligand (RANKL).

The osteogenic medium was the complete culture medium supplemented with 10 nM dexamethasone(Solarbio), 10 mM β-glycerophosphate (Solarbio), and 50 mg/l ascorbic acid (Solarbio).

Cell(Osteogenic;50 mg/l ascorbic acid):
Osteogenic induced medium contained complete culture medium supplemented with 100 nM dexamethasone (Solarbio, Beijing, China), 10 mM β-glycerophosphate (Solarbio) and 50 mg/l ascorbic acid (Solarbio). The medium was replaced every other day. Alizarin Red–positive mineralized matrix was used to evaluate osteogenic capacity by Alizarin red (AR) staining.

In Vivo:
Rat(雄性新生SD Rat;100 mg vitamin C/kg bw/d;Gavage;30d):
Forty-five male neonatal SD rats were randomly divided into five groups (n=9 each): corn oil control group, 500 mg DEHP/kg of body weight per day (bw/d) group, 500 mg DEHP/kg bw/d+200 mg vitamin E/kg bw/d group, 500 mg DEHP/kg bw/d+100 mg vitamin C/kg bw/d group, and 500 mg DEHP/kg bw/d+200 mg vitamin E/kg bw/d+100 mg vitamin C/kg bw/d group. DEHP and vitamin E (Solarbio, China) were dissolved in corn oil, and vitamin C (Solarbio,China) was dissolved in distilled water. From postnatal day (PND) 1 to 30, the male rats were administered corn oil, DEHP, vitamin C, and/or vitamin E by gavage each day according to their group. On PND 36, six rats of each groups were sacrificed and their testes were immediately isolated. The rest of rats were fed with routine feed until PND 90.
