Quantification of m6A Level at Single Base Using SELECT:

First, 1 μg total RNA were mixed with 40 nm Up Primer, 40 nm Down Primer and 5 μm dTTP in 17 μL 1×CutSmart buffer. The RNA and primers were annealed by incubating mixture at a temperature gradient: 90 ℃ for 1 min, 80 ℃ for 1 min, 70 ℃ for 1 min, 60 ℃ for 1 min, 50 ℃ for 1 min, and then 40 ℃ for 6 min. Subsequently, a 3 μL of mixture containing 0.01 U Bst 2.0 DNA polymerase, 0.5 U SplintR ligase and 10 nmol ATP (Solarbio, IA0590) were added in the former mixture to the final volume 20 μL. The final reaction mixture was incubated at 40 ℃ for 20 min, denatured at 80 ℃ for 20 min and kept at 4 ℃. Afterward, quantitative real-time PCR (qPCR) reaction was amplified on a CFX-96 Real-Time System.

Construction of biosensor for protein kinases detection:

Scheme 1B presents the ECL biosensor for protein kinases detection based on the gC3N4-MXenes nanoprobe. The cysteine-terminated kemptides were assembled on the Au electrode through the Au-S bond. Then, the kemptide/Au electrode was treated with MCH to block blank binding sites. In the presence of PKA and ATP, the hydroxy in the serine of kemptide was phosphorylated and conjugated with g-C3N4-MXenes nanocomposites by the chelation interaction between Ti defect sites on MXenes and phosphate groups. The plenty of Ti defect sites on MXenes provides more active sites for the highly efficient recognition of phosphorylated kemptide and the nanoprobes. Owing to the excellent conductivity and catalytic activity, large surface area of MXene, the ECL signal could be improved obviously by catalyzing the g-C3N4 ECL reaction with the coreactor of S2O82- and promoting the electron transfer at the electrode interface. Hence, strong and stable ECL signal are obtained, which provides a sensitive tool for kinase activity evaluation.
