Alkaline phosphatase (ALP or AKP) is widely distributed in mammalian tissues. The optimum pH for its activity is 9.2-9.8. This enzyme mainly exists in active substance exchange sites (cell membranes):Such as the brush-like margin of intestinal epithelium and proximal convoluted tubule of kidney, the stationary cilia of epididymis epithelium, the capillary cholangium of liver, and the endothelium of arterioles and capillary arteries. It is also found in the endoplasmic reticulum, Golgi complex, swallowing vesicles, lysosomes of intestinal epithelium, neutral granules of neutrophils and smooth muscle cell membranes.
Solarbio Alkaline Phophatase Stain Kit (Modified Gomori Ca-CoS Method) uses metal precipitation method to display alkaline phosphatase activity, with cascading amplification effect, and can qualitatively or semi quantitatively detect the distribution of alkaline phosphatase in frozen and paraffin sections. This method is based on the natural existence of β- Sodium glycerophosphate is used as a substrate, which is hydrolyzed by enzymes to release phosphate. It is immediately captured by metal ions and converted into a precipitate, which is then sequentially replaced with a stable brown black attached lake for easy observation and long-term preservation.
