In Vitro:
Cell(CAL-27 and SCC-15 cells; 75 nM Vitamin D3, 37℃, 48h):
The cell apoptosis assay adopted flow cytometry using Annexin V-FITC/PI. According to the protocol, the cells treated with ASA (1 mM) or/and vitamin D3(75 nM) were incubated for 48 h at 37℃, and then the cells were rinsed with PBS buffer twice and centrifuged for 5 min (1000 rpm). Before tested, the cells were gently resuspended in Binding Buffer and stained with Annexin V-FITC (5 ug/ml) and PI(10 ug/ml) for 15 min in the dark. The data were analyzed using FACScan system.

In Vivo:
Rat(male,6?week?old Sprague Dawley rats;subcutaneous injection/subcutaneous injection)
Following a week acclimation period, 22 rats were divided into the arterial calcification group (experimental group; n=16) and normal arterial group (control group; n=6). The animals in the arterial calcification group were injected with vitamin K 11.5 mg/(100 g body weight/day) subcutaneously from the first day to the sixth, on the third day, subcutaneous injection of vitamin D3 (Beijing Solarbio Science& Technology Co., Ltd.; 30,000 U/(100 g body weight/day) was continued once per day until the fifth day, and warfarin 150 mg/kg was administered twice per day orally until the sixth day; and normal arterial group were injected with 0.9% saline of the same volume with the same method.

Rat(Sprague-Dawley (SD) rats aged 7–8 weeks;VitD3 (5 μg/kg body weight,every day);Gavage):
The pregnant rats were randomly divided into 4 groups, namely control group, PM2.5 group, VitD3 group, and VitD3 + PM2.5 group. The rats in the PM2.5 group received intratracheal instillation of 1.0 mg/mL COF-PM2.5 (3 mL/kg body weight, for 3 continuous days) and intragastric administration of VitD3-free vehicle (every day). Rats in the VitD3 group received intragastric administration of VitD3 (5 μg/kg body weight,every day) and intratracheal instillation of normal saline (for 3 continuous days). Rats in the VitD3 + PM2.5 group received intratracheal instillation of 1.0 mg/mL COF-PM2.5 (3 mL/kg body weight, for 3 continuous days) and intragastric administration of VitD3 (5 μg/kg body weight, every day). Rats in the control group received intratracheal instillation of normal saline (for 3 continuous days) and intragastric administration of VitD3-free vehicle (every day). The rats were anesthetized by pentobarbital and then sacrificed by cervical dislocation on GD21.
