Dexamethasone is a basic agent for osteogenesis induction, which can activate glucocorticoid receptors located on the surface of stem cells and significantly increase the activity of alkaline phosphatase. It can induce stem cells to differentiate into osteoblasts.

This product can be used for induction of osteoblasts and other cells.


Examples of using this product(for reference only)

In Vitro:

Cell(1 μM dexamethasone/adipogenic; 10 nM dexamethasone/Osteogenic):

The adipogenic medium was the complete culture medium supplemented with 1 μM dexamethasone (Solarbio), 0.2 mM indomethacin (Solarbio), 0.01 g/L insulin (Solarbio), and 0.5 mM isobutyl-methylxanthine (Solarbio).The osteogenic medium was the complete culture medium supplemented with 10 nM dexamethasone(Solarbio), 10 mM β-glycerophosphate (Solarbio), and 50 mg/l ascorbic acid (Solarbio).

Cell(Osteogenic;100 nM dexamethasone):

Osteogenic induced medium contained complete culture medium supplemented with 100 nM dexamethasone (Solarbio, Beijing, China), 10 mM β-glycerophosphate (Solarbio) and 50 mg/l ascorbic acid (Solarbio).  The medium was replaced every other day.  Alizarin Red–positive mineralized matrix was used to evaluate osteogenic capacity by Alizarin red (AR) staining.

Osteogenic differentiation assay:

Cells were cultured in the osteogenic medium which contained 90% a-MEM (BI), 10% FBS (BI), 10 nM dexamethasone (Solarbio,Beijing, China), 10 mM b-glycerophosphate (Solarbio) and 50 mg/L ascorbic acid (Solarbio).

Adipogenic differentiation assay:

Cells were cultured in the adipogenic medium which contained 90% a-MEM (BI), 10% FBS (BI), 1 mM dexamethasone (Solarbio), 0.2 mM indomethacin (Solarbio), 0.01 g/L insulin (Solarbio) and 0.5 mM isobutyl-methylxanthine (Solarbio).

Cell(BMSCs and HUVECs;10 μM or 100 μM dexamethasone/DEX):

The cells were used between the third and sixth passages (p3–p6) and divided into six groups for the cell counting kit-8 (CCK-8) assay: (1) control;  (2) dexamethasone (DEX;  10 μM;  Solarbio Co., Ltd.;  concentration was selected based on previous studies);  (3) DEX + 5 μM AS-IV;  (4) DEX + 20 μM AS-IV;  (5) DEX + 50 μM AS-IV;  and (6) DEX + 100 μM AS-IV.  For alizarin red staining;  alkaline phosphatase (ALP) staining;  and apoptosis, tube formation, scratch wound and Transwell migration assays, the cells were divided into control, DEX, DEX + AS-IV and DEX + AS-IV + LY294002 groups.  DEX concentration was 10 μM for alizarin red staining;  ALP staining;  and tube formation, Transwell migration and scratch wound assays;  and 100 μM for the apoptosis assay.  AS-IV and DEX were administered simultaneously.  The concentrations of AS-IV and LY294002 were 50 and 10 μM, respectively.
