In Vitro:
E. coli(3 μg/mL erythromycin):
After purification from E. coli positive strains, the resulting plasmids, pMAD-?cheAY, were electroporated into L. monocytogenes competent cells and also in the in-frame ?cheAY mutant with the selection performed on TSA-containing erythromycin (3 μg/mL). Using erythromycin as the screening condition, the L. monocytogenes strain containing pMAD-?cheAY plasmid was subcultured continuously to realize homologous recombination.

E. coli(100/200 μg/mL erythromycin):
The culture of L. plantarum SL09 strain with plasmid pMG36e or pMG36eargG required the addition of erythromycin (Solarbio, Beijing, China), with a final concentration of 100 μg/ml, meanwhile, the E. coli DH5α strain with plasmid pMG36e or pMG36eargG required the addition of erythromycin (Solarbio, Beijing, China) with a final concentration to 200 μg/ml.
