In Vitro:
Cell(epidermal cells and skin-derived precursors (SKPs),20 nM b-estradiol):
Formation of HFS and IHFSAfter collected in isolation procedure, the epidermal cells and SKPs were mixed at ratio of 1:5e9. Then the mixture was seeded into 20 ml HF induction culture medium (DMEM/F12;10% (v/v) FBS, 1% (v/v) Insulin-transferrin-selenium,0.2 nM 3,30,5-Triiodo-L-thyronine, 20 ng/ml basic fibroblast growth factor, 2 nM hydrocortisone hemisuccinate, 2 nM testosterone, Solarbio, China; 20 nM b-estradiol, Solarbio, China; 20 nM progesterone, Solarbio, China) droplet on the lid of 100 mm dish with a total cell of 5×105 in each droplet. When the lid was put upside down, the cells sedimented at the bottom of the droplet and aggregated to form HFS after 24 h culture. The culture medium was changed every other day. After 7 days culture, the HFS was collected.

Cell(hAMSCs;1×10-6 mol/L β-estradiol;3d):
For endometrial diferentiation, when hAMSCs reached 30–40% confuence, the medium was exchanged for DMEM/F12 supplemented with 2%FBS, 10 ng/mL TGF-β1, 10 ng/mL EGF, 10 ng/mL PDGF-BB, 1× 10-6 mol/L β-estradiol (Solarbio), and endometrial conditioned medium. Uninduced cells were cultured with DMEM/F12 containing 2% FBS. After 3 days, the expression of CK7 and CK19 was detected by immunofuorescence.
