In Vitro:
Phagocytosis Assay of Hemocyte(FITC-labeled C. albicans (1×105 cells, 5 μl),37℃ for 3 h in dark ):
In brief, 15 larvae per group were inoculated with 5 μl of PBS or ZnO-NP suspensions (2 mg/kg) via the last left proleg. After incubation at 37℃ for 2 h, each larva was challenged with fluorescein isothiocyanate isomer I (FITC; Solarbio, Beijing, China)-labeled C. albicans (1×105 cells, 5 μl) and incubated at 37℃ for 3 h in dark. For preparing FITC-labeled C. albicans, C. albicans suspensions (1×109 cells/ml) were incubated with 0.1 mg/ml in a shaking incubator in the dark for 1 h and washed with PBS three times. Subsequently, 50 μl of hemolymph was obtained from each larva and diluted in a 12-well plate containing 950 μl of IPS and 0.002% PTU. The plate was gently shaken followed by standing for 15 min at room temperature. The phagocytosis of C. albicans by hemocytes was visualized using an IX73 fluorescence microscope.
