X-gal is the substrate of β-Galactosidase, which is blue when hydrolyzed. Based on this characteristic, when pUC series carrier DNA (or other carrier DNA with lacZ gene) is transformed by LACz-deficient cells as host, or transfected with M13 phage carrier DNA, if X-Gal and IPTG are added to plate culture medium, due to the α-complementality of β-galactosidase, Recombinants can be easily selected based on the presence or absence of white colonies (or plaque).
