In Vitro::

Cell(HUMSCs;1 mM testosterone):

Male germ-like cell differentiation of HUMSCsThe HUMSCs used in each experiment were from passages 3–5. The male germ-like cell differentiation medium consisted of HDMEM-F12 supplemented with 5% FBS, 50% fi ltered testicular cell conditioned medium, 2 mM all-trans retinoic acid and 1 mM testosterone (Solarbio,Beijing, China). HUMSCs were grown in differentiation medium for 7, 14, or 21 days; half of the differentiation induction medium was replaced every 3–4 days. HUMSCs of the control group were cultured in normal medium (DMEM-F12 containing 5% FBS and 2% penicillin/streptomycin).

Cell(epidermal cells and skin-derived precursors (SKPs),2 nM testosterone):

Formation of HFS and IHFSAfter collected in isolation procedure, the epidermal cells and SKPs were mixed at ratio of 1:5e9. Then the mixture was seeded into 20 ml HF induction culture medium (DMEM/F12;10% (v/v) FBS, 1% (v/v) Insulin-transferrin-selenium,0.2 nM 3,30,5-Triiodo-L-thyronine, 20 ng/ml basic fibroblast growth factor, 2 nM hydrocortisone hemisuccinate, 2 nM testosterone, Solarbio, China; 20 nM b-estradiol, Solarbio, China; 20 nM progesterone, Solarbio, China) droplet on the lid of 100 mm dish with a total cell of 5 x105 in each droplet. When the lid was put upside down, the cells sedimented at the bottom of the droplet and aggregated to form HFS after 24 h culture. The culture medium was changed every other day. After 7 days culture, the HFS was collected.
