Use:

Rapid preparation of mouse monoclonal and polyclonal antibodies with ELISA titers (Cutoff of 0.1000) of 1:10,000-1:100,000 or higher can be achieved by a 3-week, 2-dose rapid immunization program.

Note:

Transport at room temperature, store 2-8oC, sterile access, valid for 12 months. (Do not freeze!)

Usage:

1. Dilute the antigen with normal saline to 2 times the final concentration (prepared according to the dosage of 50μl per injection). Note: The recommended dosage of antigen is (1) subunit protein antigen with weak immunogenicity 5-20μg per injection; (2) Inactivated whole virus or whole bacteria and virus-like particle antigen with strong immunogenicity 1-5μg per injection; (3) The small molecule antigen coupled with the carrier protein is 20-50μg per injection.

2. Fully mix the adjuvant, take out the required dosage (50μl per injection) and rapidly mix the antigen according to the volume ratio of 1:1 under sterile conditions. Note: Precipitation of this adjuvant is a normal phenomenon, and the faster the mixed operation with antigen, the better.

3. Immunized mice were injected through the calf muscle of the hind leg, with 100μl injected into each mouse. . Note: It is normal for this adjuvant to precipitate after mixing with antigen. It should be thoroughly mixed before being pumped into the needle and injected as soon as possible after being pumped into the needle.

4. On day 14, administer 1 dose of immunization in the same way. Note: Each adjuvant and antigen is ready for use.

5. On the 21st day, a trace amount of tail blood was collected for ELISA determination, and the antibody titer should reach 1:10,000-1:100,000 or higher, and then whole blood could be collected or antigene-impact immunization and spleen cell fusion could be performed by conventional methods.

Compared with the conventional Fredrin adjuvant, SoFastAntibody immune adjuvant has many advantages, such as short immune cycle, less injection times, no need for emulsification, low amount of antigen, high antibody titer, high antibody affinity, easy to obtain conformational epitopes of antibodies, etc., detailed as follows:

1.SoFastAntibody requires only two doses of immunization, whether used for monoclonal or polyclonal antibody preparation, which can reduce immunization times compared with Fredrin adjuvant.

2.SoFastAntibody can greatly save the total amount of antigen by reducing the number of immunization shots and reducing the amount of antigen per shot. The recommended dosage of antigen is (1) 5-50μg per dose of subunit protein antigen with weak immunogenicity (usually 5-20μg for mice and 20-50μg for rabbits); (2) Inactivated whole virus or whole bacteria and virus-like particle antigen with strong immunogenicity 1-10μg per injection (usually 1-5μg for mice and 5-10μg for rabbits).

3.SoFastAntibody has fast antibody production, high antibody titer and high antibody affinity. For example, the standard immunization procedure, whether used for monoclonal or polyclonal antibody preparation, requires only two doses of immunization over three weeks, usually obtaining a high affinity antibody level of 1:10,000-1:10,000,000 with an ELISA titer (Cutoff value of 0.1000) at week 5.

4.SoFastAntibody does not destroy the natural conformation of the antigen, so it is easy to screen and obtain monoclonal antibodies against the epitope of the conformational type of antigen, which is an important feature that Fredrin adjuvant does not have.

5.SoFastAntibody is a class of water-soluble adjuvant, which does not require the complex emulsification process of Freudenstein's adjuvant when used, and the antigen and adjuvant can be used in immunizing animals simply by mixing.

6.SoFastAntibody uses muscle immune pathway, which is much easier to use than foot pad immunization or intrasplenic immunization used in the routine preparation of mouse monoclonal antibodies.

7. An important use of SoFastAntibody is that it can be easily used to produce mouse polyclonal antibodies. The conventional polyclonal antibody preparation mainly uses rabbits, which not only has many immunization times, large amount of antigen and slow antibody production, but also needs to entrust special units for preparation due to high technical requirements. With SoFastAntibody immune adjuvant, any laboratory animal can easily and quickly prepare mouse polyclonal antibodies, and the standard immunization procedure requires simply immunizing five mice to obtain 1ml of high-quality mouse polyclonal antibodies after five weeks. (This adjuvant is strongly recommended for the preparation of mouse polyclonal antibodies, and 1ml of high-quality mouse polyclonal antibodies is sufficient to meet the experimental needs of most users, including for enzym-linked immunization, western blotting, flow cytometry, immunohistochemistry and immunoprecipitation, etc.)
