In Vitro:
Briefly, bFGF and LET-bFGF were labeled with Cy7 (Solarbio, Beijing, China) by dissolving Cy7 in DMF and mixing with the recombinant protein [in phosphate-buffered saline (PBS)] at a 20:1 ratio while stirring at room temperature for 4 hours. Resulting solutions were concentrated and purified by dialysis to remove uncoupled Cy7.
Twenty-four male Sprague–Dawley rats were randomized into four groups: normal plus bFGF, normal plus LET-bFGF, irradiation plus bFGF, irradiation plus LET-bFGF. The rats in the irraidation group received whole thorax radiation with total radiation dose of 15Gy using an X-ray generator. The rats in the normal group didn’t receive thorax radiation. Next, bFGF-Cy7 and LET-bFGF-Cy7 (500 μL, 30 μM) were intravenously injected into rats immediately after the irradiation and allowed to circulate for 1 hour before imaging rats under the IVIS imaging system (Perkin Elmer, Waltham, MA) at 710 nm. Finally, the lung, heart, liver, spleen, and kidneys were dissected and imaged by the IVIS imaging system.
