insulin is a polypeptide hormone secreted by pancreatic β cells in a double-stranded form (α, β). It is composed of 51 amino acids, of which the α chain contains 21 amino acids and the β chain contains 30 amino acids. The two chains are connected by two pairs of disulfide bonds, and the alpha chain itself contains another pair of disulfide bonds. The amino-terminal portion of preproinsulin carries a 23-to 30-amino acid signal peptide that transfers preproinsulin to the endoplasmic reticulum, where it is immediately cut off to release proinsulin. Proinsulin is composed of the insulin β chain, C-peptide and α-chain. The released proinsulin is then transported to the Golgi apparatus, where it is packaged and converted into insulin, which is released into the blood together with C-peptide.
Insulin is the only known protein hormone in the body that can lower blood glucose. It is not only responsible for regulating the intake, utilization and storage of glucose, amino acids and fatty acids by cells, but also inhibits the degradation of glycogen, protein and fat. The function of insulin in different species is basically the same, but the composition of amino acids is different. The structure of human insulin is as follows:
This product is a full-length sequence of human insulin prepared by genetic recombination technology, the molecular weight is about 5800 Da, and the structure is the same as that of natural human insulin.
It is often used as an auxiliary component of cell culture. The mother liquor of 1-10mg/ ml is prepared and stored at -20℃ according to the single dose to avoid repeated freezing and thawing. In addition, the mother liquor after the removal of bacteria by filtering with a low-protein adsorption filter membrane, or with a suitable antibacterial agent such as 0.1% thiomersalate or sodium azide, can also be stored at 2-8 ℃ for up to 6 months.
