In Vitro:
Arabidopsis（14 day old，Col-0, dcp5-1 and ssf-2 seedlings；1 mM Cordycepin）:
Briefly, 14-day-old Col-0, dcp5-1 and ssf-2 seedlings were incubated in 3 mL of incubation buffer (1 mM Tris–HCl pH 7.5, 1 mM NaCl, 1 mM KCl, 15 mM sucrose) with rotation at 75 rpm in 12-well plates for 15 min. The incubation solution was then replaced with 3 mL of fresh buffer containing 1 mM cordycepin (Solarbio, Beijing, China, cat: 70-03-0). Samples incubated at 22 ℃ were harvested at different time points (0.5, 1, 2, 4, 6, and 12 h), frozen in liquid nitrogen, and subjected to total RNA isolation and RT-qPCR analysis as de_x005f scribed above.
