The correct heating method is essential for the full dissolution and coagulation of agarose. Here, we provide the following points as operational reference for the correct preparation of agarose gels.
1. The time required for the agarose to dissolve by heating is short.
2. Agarose does not dissolve in TAE or TBE buffers. When heated, agarose particles hydrate to form a solution. Hydration is time-dependent and different agarose hydration points are different. Agarose has high purity and ultrafine particles, which is very suitable for electrophoresis experiment carrier. Compared with other agaroses, agaroses hydrate quickly, and users do not need to boil for too long, otherwise it is easy to lead to thick glue, which is not conducive to mixing, and easy to break after gel.
3. Melting point range: 62-68℃, gel temperature (1.0%) : 24-29℃
Agarose gel preparation
A 1% concentration agarose gel was prepared with 1×TBE solution
1. The volume of the conical bottle used for preparing the gel should be 2-4 times the volume of the gel liquid. Slowly add agarose to the buffer, stirring while adding, to prevent agarose accumulation. Record the total weight of the bottle and solution.
2. Microwave oven on high heat for 30s and adjust the heating time according to the volume of the prepared solution. The heating time is related to microwave oven, bottle size and agarose concentration.
3. Shake the agarose solution.
4. Heat it again for 30s on high heat, and shake the agarose solution.
5. Put the solution back in the microwave and bring to a boil on high (about 10-35s). Agarose gel may boil violently when moving. Use caution to avoid burns. Remove from the microwave and let cool at room temperature for 1-2min, shaking gently to allow bubbles to escape.
6. Put it back in the microwave oven, heat it on high heat, let it boil for about 15s, observe the dissolved state of agarose, if there are still particles, repeat this step until all crystals are dissolved.
7. After the agarose is completely dissolved into liquid, re-weigh the total weight, calculate the amount of evaporated liquid, fill up with water to the original weight, shake the liquid well.
8. It is recommended that the glue is poured when the liquid is cooled to 50-55℃, which is conducive to the uniform gel aperture and will not damage the glue machine. Gently shake the agarose solution before gluing to release the remaining bubbles in the gel solution.
9. Pour glue into the glue making tank, the thickness of the general glue is 3-5mm, and try to eliminate the bubbles between the comb holes or at the bottom after the glue filling.
10. Place at room temperature (30-45min) to fully set the glue.
If you want to prepare different concentrations and different volumes of gel, please note the following two points:
1. Shake the gel solution at least 2 times before boiling.
2. After boiling, observe the degree of solution every 10-15s (the interval depends on the concentration and volume of the glue).
