Measurement significance:
Starch is the main storage form of sugar in plants, and its content determination is important for evaluating the nutritional value of food and investigating the sugar metabolism in plants.
Determination principle:
80% ethanol can be used to separate soluble sugar and starch in the sample, further acid hydrolysis is used to decompose starch into glucose, and anthrone colorimetry is used to determine the glucose content, which can be used to calculate the starch content.
Reagents and Equipments Required but Not Provided:
Visible spectrophotometer/enzymometer, water bath, adjustable pipette, micro quartz cuvette/96-well plate, mortar and pestle, ice, concentrated sulfuric acid (express delivery is not allowed), mortar and pestle and distilled water
