In Vitro | 阿魏酸对FGFR1表现出很强的抑制活性,在1μM时抑制率为92%。在阿魏酸处理5至40μM,持续24小时后,FGF1刺激的HUVEC增殖显著降低。阿魏酸在高达20μM时不会发挥显著的细胞活力,但超过30μM阿魏酸与对照相比在HUVEC中表现出细胞毒性作用。阿魏酸以剂量依赖性方式抑制FGF1诱导的HUVEC迁移和侵袭。阿魏酸显著抑制FGF1诱导的PI3K和Akt的磷酸化。阿魏酸处理显著抑制FGF1刺激的MMP-2和MMP-9表达[1]。 |
In Vivo | 用阿魏酸(FA)治疗有效抑制FGF1诱导的新血管形成。发现与用DMSO处理的对应物相比,胃酸给予阿魏酸显著抑制肿瘤体积和肿瘤重量。此外,阿魏酸治疗耐受性良好,载体组和FA治疗组之间的体重没有显著差异[1]。通过口服途径给予的阿魏酸(0.01,0.1,1或10mg/kg)显著降低了强迫游泳试验(FST)和尾部悬吊试验(TST)中的不动时间,而在开放场试验中没有产生影响。结果表明阿魏酸(0.001 mg/kg,po)的施用增强了氟西汀(5 mg/kg,po)在TST中的抗抑郁样作用[2]。 |
Cell Experiment | 将HUVEC(5×10 4个细胞/孔)接种到凝胶化的24孔培养板上,并在含有15%FBS的ECGS中培养。用DMSO(0.1%)或不同浓度的阿魏酸(FA)(0,2.5,5,10,20,30,40μM)处理HUVEC 24小时。通过MTT测定确定细胞活力。孵育4小时后,用酶标仪在450nm处测量吸光度。结果由每个实验的六个重复计算。进行了三次独立实验[1]。 |
