Cell Experiment | 以对数期计数A549细胞,并将6000个细胞(90μL体积)置于96孔板中。将10μL不同浓度的丹参酮IIA(终浓度80,60,40,30,20,15,10,5和2.5μM)和ADM(最终浓度8,4,2,1,0.5和0.25μM)加入到药物组,而阴性对照组(载体组)仅加入10μLDMSO或不含丹参酮IIA或ADM的生理盐水。将细胞与CCK-8试剂(100μL/ mL培养基)再孵育2小时,并使用酶标仪在450nm处读取吸光度。细胞增殖抑制率根据下式计算:增殖抑制率(%)= 1 - [(A1-A4)/(A2-A3)]×100,其中,A1是药物实验组的OD值,A2是空白对照组的OD值,A3是不含细胞的RPMI1640培养基的OD值,A4是与A1浓度相同但没有细胞的药物的OD值。 |
