The T7pLysY strain is an enhanced strain of E. coli BL21, deficient in Lon and OmpT proteases, and is used for the expression of toxic or non-toxic proteins. The advantage of this strain compared with BL21(DE3)pLysS and BL21(DE3)pLysE strains is that the T7 RNA polymerase gene is integrated in the lac operon region of the bacterial chromosome, and there is no pre-λ-phage sequence in the genome. LysY expressed T7 lysozyme retained the inhibition effect on T7 RNA polymerase but lost the amidase activity of hydrolyzing cell wall, which was conducive to reducing the background expression of genes and avoiding the cleavage of bacteria during induction, and the strain was resistant to T1 phage infection. T7pLysY expression receptor cells were prepared by special technology, and the conversion efficiency of pUC19 plasmid was greater than 108 cfu/μg.
