It is often used in combination with LPS to establish pancreatitis models. The combined administration of hyraxin and LPS can make up for the defects of single hyraxin modeling to some extent. As an endotoxin, LPS activates monocytes to release cytokines that initiate systemic inflammatory responses. Hyranin stimulates pancreatic enzymes to destroy the pancreas and continuously activates inflammatory cells to release inflammatory factors. LPS subsequently disrupts the normal response of inflammatory mediators, and then develops local pancreatitis into systemic severe inflammatory response.

In Vitro:
Cell (Rat pancreatic acinar cell line AR42J; 100 nmol/l caerulein,24h):
Rat pancreatic acinar cell line AR42J was maintained in DMEM supplemented with 10% fetal bovine serum at 37 ℃ in a humidified atmosphere of 5% CO2. AR42J cells were treated with AMPK inhibitor(Compound C, 10 μM) or AMPK activator(Metformin, 10 Mm) for 24 h. When the cell density reached 80%, AP model In Vitro was induced by addition of 100 nmol/l caerulein (Solarbio, China) into the medium according to previous study. After 24 h incubating with caerulein, the cell was harvested.
