FURA-2AM is a high affinity calcium ion fluorescent probe that is permeable to cells. FURA-2AM is an ester form of FURA-2, a highly polar acidic compound that cannot enter living cells, and the addition of acetoxymethyl (AM) to its negative group significantly increases cell permeability. After penetrating the cell membrane into living cells, FURA-2AM is de-esterified by intracellular esterase into FURA-2 free acid, which binds specifically to cytoplasmic free Ca2+ (binding ratio 1:1). When FURA-2 binds to free Ca2+ in the cytoplasm, it exhibits an absorption shift, with the excitation wavelength peak changing from 380 nm to 340 nm, while the emission peak near 510 nm remains unchanged. There was a quantitative relationship between fluorescence intensity and the concentration of bound Ca2+. Fura-2 is generally excited at 340 nm (calcium ion -Fura 2) and 380nm (Fura 2) wavelengths, and the intracellular calcium ion concentration is calculated by using the ratio of fluorescence intensity corresponding to the two excitations. Ratio measurements minimize the effects of photobleaching in the mixture, leakage of fluorescent probes, uneven loading of probes and differences in cell thickness, providing more reliable and reproducible results.
