Anti-Flag Affinity Gel consists of a high quality murine Flag antibody covalently coupled to agarose and binds to flag-labeled proteins in conventional protein expression systems, such as bacteria, yeast, or mammalian cells. It is thus used for purification or Immunoprecipitation (IP) of fusion proteins or protein complexes with Flag tags.
Flag tags (flag-tag), Myc tags (MYC-tag), HA tags (ha-tag), His tags (HIS-tag) and GST tags (GST-tag) are the most common tags on commercial expression vectors. The expression of such tags can be used to detect the expression and function of the target protein or to purify the fusion protein. Flag-tag is a polypeptide composed of 8 amino acid residues (DYKDDDDK), commonly used in the form of Flag and 3X Flag. Through genetic engineering technology, the nucleic acid sequence of FLAG-tag is connected to the N-terminal or C-terminal of the target gene. Flag-tag has the following advantages: Flag-tag usually does not interact with the target protein and does not affect the function and properties of the target protein, which is conducive to the downstream study of fusion protein; Flag-tag is used as the tag protein, and the expression, location and function of the target gene can be detected by Flag antibody or Anti-Flag affinity gel, or the target protein can be purified and immunoprecipitated. The Flag fused in the N-terminal can be excised by Enterokinase (EK) (DDDK) to obtain specific target proteins. Based on the above advantages, Flag tags have been widely used in protein expression, purification, identification, functional research and protein interaction and other related fields.
Anti-Flag Affinity Gel (also known as Anti-DYKDDDDK Affinity Gel/Beads/Resin) specifically binds to the Flag fusion protein, It is widely used in the purification or immunoprecipitation experiments of fusion protein or protein complex with Flag label.
This product has a high settled protein binding amount and strong specificity: settled gel contains about 8mg of Flag antibody per ml and can bind about 1mg of settled protein with strong specificity and few non-specific heteroproteins.
This product can be combined with various forms of Flag label proteins: This product specifically binds to methionine-modified N-terminal Flag fusion proteins (Met-Flag-Protein), N-terminal Flag fusion proteins (flag-protein), C-terminal Flag fusion proteins (Protein-Flag), or fusion proteins with a Flag in the middle.
Various elution methods are available for this product:
This product can be eluted by a variety of elution methods, including Flag peptides, acidic, alkaline, neutral and SDS-PAGE loading buffers, depending on the structural integrity, biological function and subsequent application requirements. In particular, the light chain and heavy chain that do not contain antibodies after elution of Flag polypeptide can effectively solve the interference problem of light chain and heavy chain in Western experiments after immunoprecipitation.
This product can be reused many times and is cost-effective: under normal circumstances, this product can be recycled 4-10 times when used for the purification of the same protein. If used for co-immunoprecipitation to detect protein-protein interactions, reuse is not recommended.
This product is easy to use: It is stored in TBS buffer and does not contain glycerin, which does not need to be removed during use, and it is very easy to mix and absorb, and the absorption loss is also less.
