CSPs (5.0 mg) was dissolved in 1.0 mL pure water and injected into the Shodex OH pak SB-804 column (300 mm × 8.0 mm i.d.). The flow rate of the mobile phase was 0.8 mL/min. The injection volume was 20 μL. T-series dextran was used as a standard.

Ion chromatography was employed to determine the monosaccharide composition of the PCP-100, and it was coupled to a column (150 mm × 3 mm) and a pulsed amperometric detector.The PCP-100 (1 mg) was hydrolyzed by adding trifluoroacetic acid (2 M, 1 mL) for 3 h at 110 ℃. Following drying under nitrogen, the hydrolysate was mixed thoroughly with methanol to completely remove the excess trifluoroacetic acid. Subsequently, the hydrolysate was diluted with ultrapure water, and the injection volume was 1 mL. The mobile phase consisted of two solvents, in which mobile phase A was sodium hydroxide and mobile phase B was sodium acetate in sodium hydroxide. The flow rate was set at 0.4 mL/min and the column temperature was set at 30 ℃.

GC
The six tea polysaccharides were hydrolyzed in 2 mol/L trifluoroacetic acid at 100 ℃ for 6 hr, respectively . The hydrolysate was reduced by NaBH4 and acetylation with acetic anhydride, converting into its respective alditol acetate. The derivative was analyzed by a GC (GC-7900, Shanghai, China) with a capillary column (HP-5, 30.0 m × 0.32 nm × 0.5 mm). The temperatures of injector and detector were 250 and 280 ℃, respectively . N2 was used as the carrier gas and maintained at 1.0 mL/min.

A Shimadzu LC-20AT chromatography unit equipped with a Shodex OH pak SB-804 column (8 × 300 mm i.d., YMC Co., Ltd, Kyoto, Japan) and a refractive index detector (Shimadzu RID-10 A) was employed in these studies.Briefly, pectin (5 mg) was dissolved in distilled water (1 mL) and injected into the column. Distilled water was used as the mobile phase, and the flow rate was 0.8 mL/min. The column temperature was maintained at 30 ℃. The injection volume was 20 μL.

HPLC
HPLC analysis was performed on an LC20A HPLC system (Shimadzu, Japan) equipped with an SPD-20A ultraviolet detector and a C18 column (250 mm×4.6 mm, 5 μm, Shimadzu, Japan). The mobile phase was a mixture of 0.1 mol/L NaH2PO4-Na2HPO4 buffer (pH 6.7) and acetonitrile (83:17), and a flow rate of 1.0 mL/min was used. The wavelength of detection was 245 nm, and the column temperature was 30?C.
