The cell wall of acid fast bacteria (such as Mycobacterium, Mycobacterium tuberculosis, Nocardia, etc.) contains a large amount of lipids surrounding the outside of peptidoglycans, so acid fast bacteria are not colored or lightly colored in general bacterial staining solution. The traditional dyeing method requires heating and extending the dyeing time to promote its coloring. Once the branching acid in the cell wall of acid fast bacteria combines with dyes, it is difficult to be decolorized by acidic decolorization solution, hence the name acid-fast staining. The most representative one is the Ziehl-Neelsen staining method for Mycobacterium tuberculosis, which is recommended by WHO for thermal staining.
The Acid-Fast Bacillus(AFB)Stain Kit(Auramine O Method) belongs to the acid fast bacterial fluorescence staining method, which does not require heating, is simple and safe to operate. The staining principle is to use Auramine O staining and double staining under room temperature conditions, and then check the result with a fluorescence microscope containing ultraviolet light. Acid fast bacteria appear bright yellow, while other bacteria and substances in the background appear dark yellow. This method can be used for low magnification microscopy, so it can quickly identify acid fast bacteria.
