The cell wall of acid fast bacteria (such as Mycobacterium, Mycobacterium tuberculosis, Nocardia, etc.) contains a large amount of lipids surrounding the outside of peptidoglycans, so acid fast bacteria are not colored or lightly colored in general bacterial staining solution. The traditional dyeing method requires heating and extending the dyeing time to promote its coloring. Once the branching acid in the cell wall of acid fast bacteria combines with dyes, it is difficult to be decolorized by acidic decolorization solution, hence the name acid-fast staining.The most representative one is the Ziehl-Neelsen staining method for Mycobacterium tuberculosis, which is recommended by WHO for thermal staining.

The Acid-Fast Bacillus(AFB)Stain Kit(Ziehl-Neelsen's Method) belongs to the acid fast bacterial thermal staining method, which is fast and easy to operate. The dyeing principle is that under heating conditions, the fuchsin staining solution better penetrates the cell wall of acid-fast bacteria for coloring. After re-dyeing with methylene blue, the acid fast bacteria still turn red, while other bacteria and substances in the background turn blue. This staining solution has a deeper staining depth and clearer contrast compared to Kinyoung's staining method.
