Hemosiderin is a kind of hemoglobin derived pigment, which is golden yellow or brown yellow particles. Because it contains iron and is golden yellow, it is called hemosiderin. When red blood cells are engulfed by macrophages, hemoglobin is broken down into an iron-free orange hemosiderin and an iron-containing hemosiderin by the action of lysosomal enzymes. Prussian blue reaction (Perls) is also known as hemosiderin staining, that is, hemosiderin can produce blue after treatment with potassium ferrocyanide and dilute acid. It is commonly found in phagocytic cells or interstitium, and mainly shows trivalent iron salts. Therefore, the Lillie method is also a good method to show ferric iron, and its specificity is better.
The kit is mainly composed of Potassium Ferrocyanide and nuclear fast red, ferric iron is dark Prussian blue, and the nucleus is red.
