There are various methods for staining mucus material, such as AB-PAS staining, mucus HID-AB staining, and standard Alisin blue staining. Most of the above methods utilize the principle that cationic dyes can bind to acidic groups, i.e., Alisin Blue forms insoluble complexes with anionic groups such as carboxyl and sulfate contained in the tissue.
Mucin Heterochromatic Staining Solution (Azure A method) uses the heterochromatic property of azure, after staining, the color of the tissue or tissue components stained with the original color of the dye complex is obviously different from the original color of the dye complex and can form a sharp contrast. After staining, the treatment of the section is very important for the stability of the heterochromatic staining and should be preserved in ethanol.
