5(6)-CFDA is a live cell fluorescent tracer probe that is permeable to the cell membrane and thus can be loaded into the cell for incubation. Once inside the cell, the non-fluorescent CFDA is hydrolyzed by lactase into carboxyfluorescein, producing fluorescence. These fluorescent products can only accumulate in cells with intact cell membranes. Therefore, dead cells without intact membranes cannot be stained. CFDA labeling can be used to detect cells using fluorescence microscopy or flow cytometry.
