Fluo-5N, an analogue of Fluo-4, has a low calcium-binding affinity and is suitable for detecting intracellular calcium levels in the range of 1μM to 1 mM. Fluo-5NAM can be loaded directly into living cells by adding the dissolved indicator directly into a dish containing cultured cells. It is compatible with an argon ion laser source excited at 488 nm, making Fluo-5N useful for confocal microscopy, flow cytometry, and microplate screening applications. After calcium binding, its fluorescence intensity increased by > 100 times.
