DAF-FM DA is a membrane permeable nitric oxide (NO) fluorescent probe with Ex/Em=495/515nm. DAF-FM DA actively diffuses through the cell membrane, and once inside the cell, intracellular esterase deacetylates it to form DAF-FM. DAF-FM itself is very weak, but the fluorescence is enhanced by about 160 times after reaction with NO.
Procedure (for reference only)
1. Prepare live cells on cell suspension or slide;
2 Dilute the DMSO storage solution with an appropriate buffer or fresh medium (not serum and phenol red). The recommended initial working concentration range is 1-10μM.
3 The cells were hydrolyzed with the diluted DAF-FM DA working solution for 20 to 60min, and the incubation temperature was 4 to 37℃. Adherent cells do not need to be digested and reloaded.
4 Thoroughly wash cells with PBS, pH 7.4 until excess probes are removed. Replace the fresh slow medium solution or medium and incubate it for 15-30min, so that the lactase can fully de-esterify the probe.
5 The maximum excitation and emission wavelengths are 495 and 515nm, respectively. Because these wavelengths are very similar to fluorescein, individual fluorescein or FITC detection systems are suitable for each.
