Weakly fluorescent DAF-2 is a sensitive fluorescent indicator, which is commonly used to detect nitric oxide (NO). It reacts with NO in the presence of oxygen to produce highly fluorescent fluorescein. Its fluorescence can be detected using a standard FITC filter set. It can be used for real-time NO detection, but the optimal DAF-2 concentration must be experimentally determined to perform cell NO detection. For example, by subtracting DAF-2 autofluorescence from the total fluorescence measured, low DAF-2 concentrations (e.g. 0.1μM) can be used for real-time detection of nitric oxide in endothelial cells.
