Nicotinamide is a B3 vitamin that plays an important role in cellular physiology by promoting NAD + REDOX homeostasis and providing NAD + as a substrate for a class of enzymes that catalyze non-redox reactions. Nicotinamide is an indispensable nutrient in organoid culture. It is commonly used in the culture of gastrointestinal tract, liver and breast organoids to promote the formation of organoids and extend the life of organoids.


Examples of using this product(for reference only)

In Vitro:

Cell(human oral keratinocytes (HOKs) and human gingival epithelial cells (HGEs);4-16 μg/ μL Nicotinamide/NAM;1h):

The cells were cultured in DMEM, 100 U/ mL of penicillin, and 100 mg/mL of streptomycin supplemented with 20% FBS in 5% CO2 at 37℃ at a density of 5,000 cells per well in 96-well cell culture plates.  The final concentrations of NAM were 4 μg/μL, 8 μg/μL, and 16 μg/μL and a medium without NAM served as negative control.  After incubation under 5% CO2 at 37℃ for 1 h, the cells were washed with sterile PBS, and 0.1 mL of the CCK-8 reagent was added to each well.  The plate was then incubated at 37℃ for 3 h. Then the absorbance was measured at 562 nm using a spectrometer.  The statistics were normalized as cell via bility, and the result of the control group was set as 100%.  Each concentration was tested in three wells, and the CCK-8 assays were performed in triplicate.

In Vivo:

Rats(Sprague-Dawley rats;16 or 32 μg/μL Nicotinamide/NAM;2 min twice daily,4 weeks):

The infected animals were randomly assigned into four groups of seven animals: (1) 32 μg/μL NAM, (2) 16 μg/μL NAM, (3) 250 μg/mL of fluoride (as positive control), and (4) ddH2O (as negative control).  Animals in each group were treated with the compounds above topically using a camel hair brush for 2 min twice daily for 4 weeks.  The rats were fed the cariogenic Keyes 2000 diet and 5% sucrose water ad libitum during the inoculation of S. mutans and the 4-week treatment.  The rats’ weights were recorded to monitor for signs of toxicity.
