Mucin Staining Kit (Mild Methylation Method) is a type of chemical modification and blocking method, which is based on the principle of eliminating the basophilicity of COOH by using an acidic alcohol solution.
Alcian staining solution at pH 2.5 ionizes the carboxyl groups within the tissue with a negative charge, forming a salt bond with the cations in Alcian blue, which stains tissues with carboxyl groups (e.g., proteoglycans/hyaluronic acid, and epithelial acidic mucins), and is primarily used to identify acidic groups in mucins. Untreated mucin sulfate, proteoglycan sulfate, salivary mucin, and hyaluronan stain blue, and treated sections stain light blue.
